Alkaline phosphatase production by Enterobacter hormaechei isolated from birds fecal waste and its optimization.

Abstract

Phosphatases producing bacterial strains (PSB) were isolated from bird’s fecal matter, five efficient phosphatase producing bacterial strains were selected on the basis of their ability to form clear zone on Pikovskaya's agar medium. The colony showing maximum zone of clearance was selected and proceed for morphological and molecular identification. Enterobacter hormachei was identified using molecular identification. The partially purified enzyme shows 34±1.4 U/ml activity at 10 mM substrate concentration, at pH 8 and on temperature 400C after 30 minutes of incubation. Phosphatase activity found to be strongly inhibited in presence of 1 mM EDTA and enhanced by PMSF. Hence Enterobacter hormaechei potential phosphatase producer and in future may serve as good bio fertilizers and best inoculum to solve environment issues like eutrophication.
