Preparation & Evaluation of Inclusion Complex of Telmisartan with Natural Flavonoids and Citric Acid

Abstract

The main aim of the present study is to Preparation & Evaluation of Inclusion Complex of
Telmisartan with Natural Flavonoids and Citric Acid. Inclusion complex of Telmisartan with
natural flavonoids and citric acid were prepared by different methods i.e. kneading methods,
physical mixing methods and solvent evaporation etc. Dissolution studies on the pure drug,
and the inclusion complexes prepared by different methods were performed in pH 7.5
phosphate buffer using USP TDT-08L dissolution test apparatus with a paddle stirrer. The
stirring speed employed was 75 rpm, and the temperature was maintained at 37°C±0.5°C.
Powdered samples of each preparation, equivalent to 40 mg of Telmisartan, were placed in
the dissolution medium. Samples were withdrawn at different time intervals, filtered and
analyzed by measuring the absorbance spectrophotometrically at 296 nm. The prepared
tablets were evaluated by various qualitative test i.e. weight variations, hardness, in-vitro
dissolution, carr’s index etc. It was also observed that, the incorporation of citric acid into
these complexes accelerated the dissolution phenomena. The tablets formulated with the
selected complexes yielded the higher dissolution rate than the marketed formulation. Thus
this investigation resulted in the development of Telmisartan tablets with superior in vitro
dissolution properties. The drugs Telmisartan exhibited pH dependent and particle size
dependent solubility. The drug readily forms complexes with luteolin and Quercetin and the
dissolution rate is influenced by the method of preparation.
